dc.description.abstract | The bacterial or vascular wilt MV, of eucalyptus, caused by R. solanacearum, is a disease that affects the production in the different phenological phases of the plant, being reported as a limitation in the production of seedlings in the nursery and also responsible for the death of trees in field. The objective of the research was to identify the causal agent of eucalyptus vascular wilt, Eucalyptus urograndis in the department of Córdoba. In the first phase of this study, samples of infected plants were taken in the field, which were placed in humid chambers for 72 hours and the exudate that appeared from the samples was sown in semi-selective TZC and SMSA media. Six isolates with watery-looking and reddish colonies were obtained, they were purified and KOH, Gram stain and oxidase tests were performed. These were subjected to a hypersensitivity test in non-host tobacco plants (Nicotina tabacum) by inoculating them with a concentration of 108 cfu / ml of R. solanacearum, in the same way pathogenicity tests were carried out in the host plant E. urograndis, clone 14 susceptible to MV. For each isolate, five plants were inoculated and the control was inoculated with distilled water. The inoculation was carried out on the stem 5 cm above its base, depositing 0.5 ml of the bacterial solution at the previously evaluated concentration (108ufc / ml), then the wound was covered with cotton and sealed with parafilm. Destructive sampling was carried out at 104 days post inoculation, to describe external and internal symptoms, of the six isolates, two caused pathogenicity, in eucalyptus and hypersensitivity in tobacco, these two isolates were characterized by PCR and identified as strain 1 and strain 2. A second experiment was established where the resistance of 14 clones of the cultivar E. urograndis susceptible to vascular wilt was determined, taking as reference clone 14. For each clone there were five plants of which two were inoculated with the strain 1, two with strain 2 and one plant with distilled water as a control. The inoculation was carried out with the methodology and concentration described above. The monitoring of symptoms in the different clones was carried out every two days for 50 days; finally, 54 days after inoculation, a destructive sampling was carried out, the external and internal symptoms of the inoculated stems were described, evaluating the vascular infection. The presence of bacterial vascular involvement was determined by the flow test in a test tube with water. Finally, to verify the presence of the inoculated bacteria, fragments of the stems with symptoms were placed in humid chambers, the bacterial exudate produced was seeded in the TZC medium and the respective Gram and oxidase tests were carried out. A qualitative analysis was carried out comparing microbiological and molecular characteristics of the isolates, in addition the degree of damage was evaluated using the proposed scale, which allowed classifying the level of damage caused by the six strains in clone 14. In conclusion, the capacity of the six isolates to cause vascular damage in clone 14, highly susceptible and two of them were identified as strains by means of the PCR technique as R. solanacearum, which were evaluated in 14 clones, where all showed susceptibility to vascular wilt , concluding that the phytosanitary risk that this bacterium represents in eucalyptus, which must be taken into account when implementing disease management plans. | eng |

dc.description.resumen | La marchitez bacteriana o vascular MV, del eucalipto, ocasionada por Ralstonia solanacearum, es una enfermedad que afecta la producción en las diferentes fases fenológicas de la planta, siendo reportada como una limitante en la producción de plántulas en vivero y también responsable de la muerte de los árboles en campo. El objetivo de la investigación fue identificar el agente causal de la marchitez vascular del eucalipto, Eucalyptus urograndis en el departamento de Córdoba. En la primera fase de este estudio se tomaron muestras de plantas infectadas en campo, las cuales se colocaron en cámaras húmedas durante 72 horas y el exudado que se presentó de las muestras se sembró en medios semi selectivo TZC y SMSA. Se obtuvieron seis aislados con colonias de aspecto acuoso y color rojizo se purificaron y se les realizó pruebas de KOH, tinción Gram y oxidasa. Estos fueron sometidos a una prueba de hipersensibilidad en plantas no huésped de tabaco (Nicotina tabacum) inoculándolas con una concentración de 108 ufc/ml de R. solanacearum, de igual manera se realizaron pruebas de patogenicidad, en la planta huésped E. urograndis, clon 14 susceptible a MV. Para cada aislado, se inocularon cinco plantas y el control se inoculo con agua destilada. La inoculación se realizó en el tallo a 5 cm por encima de su base, depositando 0,5 ml de la solución bacteriana a la concentración previamente evaluada (108 ufc/ml), luego la herida fue cubierta con algodón y sellada con parafilm. Se realizó un muestreo destructivo a los 104 días post inoculación, para descripción de síntomas externos e internos, de los seis aislamientos dos causaron patogenicidad, en eucalipto e hipersensibilidad en tabaco, a estos dos aislamientos se les realizo caracterizaron mediante PCR y se identificaron como cepa 1 y cepa 2. Se estableció un segundo experimento donde se determinó la resistencia de 14 clones del cultivar E. urograndis susceptible a la marchitez vascular, tomando como referencia al clon 14. Para cada clon hubo cinco plantas de las cuales dos se inocularon con la cepa 1, dos con la cepa 2 y una planta con agua destilada como testigo. La inoculación se realizó con la metodología y concentración descrita anteriormente. El monitoreo de los síntomas en los diferentes clones se realizó cada dos días durante 50 días; finalmente a los 54 días después de la inoculación se realizó un muestreo destructivo, se describieron los síntomas externos e internos de los tallos inoculados, evaluando la infección vascular. La presencia de la afectación vascular bacteriana se determinó mediante la prueba de flujo en un tubo de ensayo con agua. Por último para verificar la presencia de la bacteria inoculada, fragmentos de los tallos con síntomas se colocaron en cámaras húmedas, el exudado bacteriano producido se sembró en el medio TZC y se hicieron las respectivas pruebas de Gram y de oxidasa. Se realizó un análisis cualitativo comparando características microbiológicas y moleculares de los aislados, además se evaluó el grado de daño mediante la escala propuesta, lo que permitió clasificar el nivel de daño ocasionado por las seis cepas en el clon 14. En conclusión, se verificó la capacidad de los seis aislados de causar daños vasculares en el clon 14, altamente susceptible y dos de ellos fueron identificados como cepas mediante la técnica de PCR como R. solanacearum, las cuales fueron evaluadas en 14 clones, donde todos mostraron susceptibilidad a la marchitez vascular, concluyendo que el riesgo fitosanitario que representa esta bacteria en eucalipto, que debe tenerse en cuenta al implementar planes de manejo de la enfermedad. | spa |
